Antiswitch repressing production of EYFP ("Off" switch)
The presence of theophylline prevents translation of mRNA transcribed from Biobrick BBa_E0032. The absence of theophylline allows expression of the coding region of BBa_E0032.
Usage and Biology
The antiswitch acts as an allosteric riboregulator, that in the presence of theophylline prevents translation of a target mRNA by attaching 3 bp upstream of the start codon of the target sequence, inhibiting translation of the coding region. In the absence of theophylline, the antiswitch remains free from the mRNA and translation can proceed as normal. The target sequence for this antiswitch is: uagauggugagcaag. The plasmid containing this part is Blue Heron pUCminusMCS, which itself also expresses ampicillin resistance.
